Played with a standard 52-card deck plus a joker, 500 Rum involves forming sets (same rank) and sequences (consecutive cards of the same suit) to score points. The number of cards dealt depends on the number of players (13 in a two-player game, 7 in three- or four-player games). The joker acts as a wild card.

Gameplay involves drawing cards from either the stockpile or the discard pile (but you can't discard the same card you just drew). Players lay down melds (sets and sequences) on the table, earning points based on card values (numbered cards are face value, J, Q, K are 10 points each, A is 15 points, and the joker takes the value of the card it replaces). Points are also awarded for laying off cards onto existing melds.

Rounds continue until a player reaches a score of 500 or more. The final score is calculated by adding meld and layoff points, then subtracting the value of unmelded cards. Ties result in an additional round to determine the winner.
